Assembling of Palm Oil Flowering Genetic Systems

ESTATE CROPS INNOVATION- Assembling of Palm Oil Flowering Genetic Systems is one of the technological innovations generated by Indonesian Center for Estate Crops Research and Development in 2011.

In the flowering cycle of palm oil, the sexual differentiation process begins with the formation of primordial interest of the flower meristem tissue. Once it happens the sexual differentiation, namely the primordial flowers, develops into male or female flowers depend on environmental conditions.

In many flowering plants, flowering process from the beginning until the fruit is mainly controlled at the genetic level by a group of MADSBOX genes. In many species of plants, MADSBOX has a conserved structure and function (highly conserved).

There are at least three genes involved in flowering MADSBOX palm those are EgSQUA1, EgAG and EgAGL. At least one of the three genes is strongly suspected of a role also in the process of sexual differentiation in flowering palm.

In previous studies, construct genetics of PEgAG2::GFP and PEgAGL2::GFP have been assembled using Gateway technology (from Invitrogen). In addition to this, it has been identified local sources of potentially large bio regulator that can improve both the vegetative and generative growth of monocot plants, especially for palm oil.

The study consists of two main activities, namely, (1) continuation of the assembly and analysis of genetic systems in plants, and (2) further exploration bio regulator from local sources. The first activity will confirm the previous construction obtained and regenerate plants’ culture carrying the construct of PEgAG2 :: GFP and GFP :: PEgAGL2. The second activity will inventor bio regulator inducing flowering plants, which are easiest to get and use it.

Assembling and analyzing of the genetic system is a genetic construction of PEgAG2 :: GFP and GFP :: PEgAGL2 which each of them has succeeded to be sub cloning into Agrobacterium tumefacient. Both constructions have also been successfully transformed into a model of in vitro explants of tobacco plants.

Tobacco plants (plantlets) carrying the construct has also been successfully regenerated. At a given MS salts containing 0.5 ppm BAP and 30-40 g / L sucrose, regenerated plantlets shows different structure with the positive control plantlets, which are not transformed plantlets and regenerated on standard media.

This phenomenon indicates that both reporter gene constructs are expressed under conditions in vitro, or a genetic system to function properly assembled. On upland rice trial conducted in the greenhouse, bio regulator (materials) is naturally able to increase vegetative growth and number of tillers, so that productivity and quality of seeds also increased.
