ESTATE CROPS INNOVATION – In 2011, Indonesian Center for Estate Crops Research and Development, Ministry of Agriculture released innovation technology potential to multiply the seeds quickly, voluminously, and uniformly through tissue culture technology.
Provision of sugarcane seed through tissue culture follows four important stages, namely callus induction, proliferation, differentiation, and regeneration to form plantlet. For such needs, it is required to conduct a research to get micro-propagation technological package in order to meet superior variety of sugarcane which is cheap, quick, and well adapted in a wide scale.
Multiplication activities of sugarcane resulted in the use media for callus induction with addition of 2,4-D induced the calli derived from young leaves of sugar cane explants. Increasing concentrations of 2,4-D up to 3 mg/l in a medium without other ZPT addition tended to decrease the amount of explants with callus. Addition of casein hidrolisat on callus induction media did not influence number of callus production, however strongly influenced on callus quality.
To regenerate the callus to become plantlets, it was required different media formulations for each variety, however, the use auxin (NAA and IBA) on rooting medium induced the formation of roots. The multiplication was already applied to produce sugar cane seeds massively. There were 100,000 planlets of sugarcane seeds resulted from tissue culture process in FY 2011, and potential to produce 2,800,000 plantlet Budset of G2 by the end of 2012.
